Is it possible to sort a gallery by thumbs ups? (or thumbs downs)?

A gallery can't be sorted by popularity. Sorry.
You would have to go to your popular photos section on the homepage. This section would be in order of the photos popularity. The first photos would be the most popular ones.

To reset the photorank of a photo, you could replace the photo using the 'replace tool' from the tools button.
Or you could move it into a gallery where photorank is disabled. That would remove it from your popular section as well.
