Output Sharpening for Bay Photo?

I have "PhotoKit Sharpener" which allows me to set and output sharpening resolution for inkjet printers, which I assume Bay Photo uses. Do I need to do this or will Bay Photo output sharpen based on the size and material being printed on?

    Don't assume - most prints are true-photographic silver-halide process :) Some (like giclee and canvas) are ink-jet.

I read that with PhotoKit Sharpener 300ppi is the preferred resolution for printers at BayPhoto... my question is, if the file size is roughly 12x18 at 300ppi will this file be sufficient to make 30x40 prints with this level of sharpening too?

    DPI is really meaningless -- leave your photo in its' original resolution. If you crop, crop without changing the native DPI.
