Where should I start to shoot this close better?

Well I rather like it as is - lovely colours and exposure and the composition is good , the main problem for me is I can't tell what it is. Guessing it's a flower stigma or a snails eye .
Depends a lot whether you were trying to do an abstract shot of the subject or a recognisable shot .
If you were trying for something more recognisable then you have run into the main problem with macro - very narrow DOF and it looks as if you were shooting with the aperture wide open. To get reasonable DOF around 1:1 magnification which is what you were getting with that combo you need to be shooting around F11.
