Smugmug had an issue with the site in read-only mode (no uploads allowed) for a little while today in case that's when it happened: http://status.blogs.smugmug.com/.
I just ran a quick test using the plugin as I need it later tonight and it did OK. BUT, I have the latest version of the plugin (20100812.174) as well as upgraded to LR3.2 Release Candidate which was not because of any issues with the Friedl plugin but because of the plethora of bugs in the production release of LR3. If you have updated to the latest version of the plugin and are still having issue I would try installing the LR3.2RC because this fault might be with LR3 itself.
EDIT: I should add I am running LR3.2 64-bit under Win7 x64 just in case it comes to matter.
gaaaaaahhhh!! hehehehe...I hear ya...I think this one is on Adobe though. I would sometimes get probs with the plugin under the production release of LR3 so I would say to try the LR3.2 RC and re-install the plugin as well. If Jeffry was around I am sure he would off more on target advice but sands that it's the direction I would move first rather than any other work-arounds.
These days during large uploads I wonder about ISP bandwidth throttling during long/large uploads. It is especially true of the larger ISP's like Comcrap, Verizon and other providers who lock down anyone who routinely uploads large files...I am lucky because I don't have a lot of images to upload at any given time, plus my ISP is old-school and knows me well after over a decade with them...heck, I actually get calls back from their tech support when needed or just to make sure an issue is fixed. So don't discount the ISP as a factor. But still my money is on LR as a problem, potentially anyway.

Am having exactly the same problems as the OP, AND I am now using both the latest version of the Plugin AND 3.2RC. Problem is on both Publish and Export... am at end of tether as the plugin plugin promises sooo much.
Any help/advice welcome will watch this thread with interest!
I've been using it too but only the export part. The publish kept coming back with errors. I wrote to the author and, after several exchanges, this is what he suggested:
Something is definately wrong with your system or your Lightroom install... that all sounds very strange.
Consider flushing your preferences, and doing an integrety check on your catalog. And keep good backups of your catalog, in case the corruption takes a turn for the worst. Jeffrey
Since the publish part was not that important to me, I told him I'd try the re-install if and when I need to use it. I really don't think anything is wrong with my system as everything else works flawlessly. I also have the latest plugin version and LR 3.2 installed, Win 7.
Edit: Not sure why, but I went back to lightroom to give it another try and it worked fine. I just did one picture, I'll try multiple later.
I am using LR3 on a Macbook Pro. I have mainly been using the Export feature but tried the Publish feature earlier today and it worked fine.
Basically, I just entered the appropriate login credentials and setup other parameters. It work without and errors. I installed the plugin according to the information at http://regex.info/blog/lightroom-goodies/smugmug.
I say just reinstall LR and the plugin and try again.
