The 70-200 is quite a weight too, so I know what your talking about.
The 400mm Prime is like half the weight of the 70-200.

I used the 70-200 IS L 2.8 all last season and got used to the weight of it, no big deal after a while. This thing is totally different and after 8 hours my wrist was shaking. Definately need to do some working out.
Kart racing and Super Motard practice this weekend and another track day on Friday before it is "show time" with the first local race of the season. Hopefully I learn a trick or muscle up in time otherwise on April 2nd after all that use my left arm will be rubber.

It is a great lens. I found it hard to use in a strong wind though. The "trick" to using a lens this heavy and big is making sure you don't use the wrong muscles. If you get your deltoids involved you will tire out very rapidly as these are small muscles. Make sure you are not scrunching up your shoulders to support the lens. Shoulders down and back in a neutral place. Use only your arms. As per wrist get in the habit of doing wrist curls.
I don't know if it is comparable to the 300 f4 since I never used it. I went with the 2.8 due to the rave reviews and so I could connect my 1.4x TC to it to get me some more length. The 400 IS L 2.8 is impossible to hand hold over a duration and nearly double the price.
I use the 400 2.8 with and without a 2x tele and with and without a monopod. Very heavy but after several hours everything else feels like a toy in comparison. Definately worth the effort
